Filter Spam before it hits the inbox

Are there any recommendations on anti-spam techniques that happeneds before it hits the users mailbox?

Looking at Greylisting, spam assasin etc.. but does the SA filter before the users inbox? Or do they just get each message that is spam just get tagged with ~*~ ?

Any recommendations on these spam techniques BESIDES RBL would be great, as this is not working that great for us and only gets about 50% of the spam.
