If the recipient is a mailserver that rejects your 1st message with error 45x, but allows subsequent messages after a period of time, then likely need to add this to the bottom of your Exim Advanced Editor config:

This will force EXIM to retry the message after a 450 error.

Otherwise, it could be DNS issue as another poster had said. You'd need to check your exim log for the error code when the original message was sent out.
