Do you mean how can you make it so when an account is created it automatically has an index page?

That's in the cpanel3-skel directory. The cpanel3-skel directory is copied over when an account is created. For root, that directory is /root/cpanel3-skel. For resellers, it is in /home/username/cpanel3-skel.

    or

Do you mean how do you set which page, (i.e. file.ext), is the default index page if someone goes to domain.com?

That's in the "IfModule mod_dir.c" section of the httpd.conf file.
