Does any know what needs to be checked and not to be checked in the Tweak Settings?

The options defined in the Tweak settings are quite simple and have explanation for each option. You can go through them and select the options on your choice. Everyone has its own choice so there is no hard and fast rule that which options should be selected. However if you are not sure about any option, please post it here so that someone can give you better understanding for it.
